I think there's a bit of difference between what's typically thought of as American and British humour. As far as I know, though, it's not different enough that an American wouldn't be able to understand a Briton's humour.
I think a lot of Americans are just turned off by British stuff.

If you have watched Monty Python and Benny Hill, Blackadder and all the wacked out kids shows, you will get an idea of british humour. And the brit version of The Office. Kind of a parallel to American humour. Personally I prefer what I grew up with, but Im loving thiry rock and snl, how i met your mother, and the one about the science nerds.
